US 1 is a major route running from Key West in Florida to the Canadian border in Maine. Prior to the construction of I-95, it was the main route along the East Coast. The section in Maine takes an indirect route around the edge of the state to reach the Canadian border near its northernmost point.

US 1 enters Maine from New Hampshire, crossing a bridge over the Piscataqua River. It runs just south of the Maine Turnpike (I-95) along the coast until Portland, where it joins I-295 for a short distance. US 1 follows the highway through the city and then splits off. It runs near I-295 through Falmouth and Freeport.

In Brunswick, US 1 turn east to follow the coast, while I-295 continues northeast toward Augusta, the state capital. There is a short expressway section between Brunswick and Bath and then again forming a semicircular bypass around Belfast. ME 3 joins US 1 along this section, and the two routes are multiplexed from here to Ellsworth. Along this section, US 1/ME 3 crosses the Penobscot River via the Penobscot Narrows Bridge, a two-lane cable-stayed bridge. This bridge is notable for having an elevator inside one of its towers leading to an observatory at the top, the highest such bridge observatory open to the public anywhere in the world. In Ellsworth, ME 3 splits off toward Mount Desert Island and Acadia National Park. US 1 continues east along the coast.

As US 1 nears the Canadian border, it turns to the north and passes through Calais, where there is access to a bridge to New Brunswick, Canada, leading to NB 1, which is distinct from US 1. Continuing north, US 1 crosses I-95 one last time in Houlton at its second to last exit in the United States. US 1 continues north through Presque Isle and Caribou to Van Buren. There, the road straight ahead crosses the St. John River to Canada, but US 1 turns northwest to remain in the United States. It runs along the river to Fort Kent, where it is joined by ME 161. After a few blocks, US 1 splits off to the north and finally crosses the river to Canada. ME 161 continues along the river on the American side to Allagash.
