Union Turnpike begins at the junction of New Hyde Park Road (CR 5B) and Marcus Avenue in New Hyde Park. It runs west as a four-lane divided road. There is a single NY 25C reassurance shield at the very beginning.

There is a traffic light at Lakeville Road (CR 11). Turn right to go north or left to go south. The Queens-Nassau county line passes through this intersection. Union Turnpike continues straight ahead into Queens. At this intersection of three routes systems (New York State, New York City, and Nassau County), the traffic light is provided by the Nassau County Department of Public Works.
